Phospholamban (PLN) regulates cardiac contractility by inhibiting the sarcoplasmic reticulum (SR) Ca²⁺-ATPase (SERCA2a) pump. In its unphosphorylated state, PLN inhibits SERCA2a-mediadted Ca²⁺ uptake into the SR, thereby reducing cardiac contractility. Phosphorylation at Ser16 and Thr17 relieves this inhibition, enhancing Ca²⁺ reuptake and improving cardiac contraction and relaxation.
